Gabriel Osho's low first-half strike looked like it would secure a fifth 1-0 win in a seven-game unbeaten run against a Watford side who failed to register an attempt on target in the first half.
And although the Hornets improved after the interval and went close through a Jeremy Ngakia header, a poor clearance allowed Allan Campbell to smash in an injury-time second for the hosts.
Luton's latest success sees them stay in fourth, just three points adrift of second-placed Sheffield United and level on points with third-placed Middlesbrough prior to Saturday's later kick-offs.
Watford's chances of creeping into the play-offs seem remote having won just once in a six-game run that has also seen three consecutive away defeats.
